IgG4 Antibodies Induced by Repeated Vaccination May Generate Immune Tolerance to the SARS-CoV-2 Spike Protein

https://doi.org/10.3390%2Fvaccines11050991

the length limitation needs to be satisfied, but the situation is not as alarming as the truncation would infer

How would you re-write it better? The original title actually seems worse to me, but if somebody can re-write it, please do so.

[Repeated Vaccination May Generate Immune Tolerance to the SARS-CoV-2 Spike Protein]

IgG4 is not crucial to understanding the content.

omitting the requirement of repeated vaccination, and specificity to spike protien, creates an impression the hypothesis applies to much wider conditions than is actually the case.

may generate is important as this communicates the idea that this is not fact, but rather, a possibility that should be investigated further.

Makes sense. I like your version better. I was afraid of editing it too much, but this is better. Can HN adminisrators change the title, please?

> as with many other vaccines, they do not produce sterilizing immunity

I wonder if the authors emphasized that as a pre-buttal to a certain political bloc. I recall a few waves of conspiratorial talking-points going around, along the lines of: "Real vaccines protect you perfectly with one dose forever, this one doesn't, wake up sheeple", etc.

Of course it's ahistorical nonsense: By that logic, most/all vaccines ever made over 200+ years were all wrongly labeled.

> The long-term consequence of this tolerance could be the establishment of a permissive state of the host leading to chronic infection and other unintended consequences induced by mRNA vaccination in susceptible individuals.

Alternately, the long-term consequence could be that the disease is still killed in a timely manner while the human is less-likely to suffer from an overreaction, like hyperinflammatory cytokine storms.

It gets a mention later in the discussion section:

> The increase in IgG4 levels [...] could reflect a tolerance mechanism that could prevent immune over-reactivity (cytokine storm) and progression to a critical stage. However, this exacerbated immune reaction does not occur in young and healthy people, and it has been documented only in older patients with genetic susceptibility and those with comorbidities.

How about this part of the paper:

However, emerging evidence suggests that the reported increase in IgG4 levels detected after repeated vaccination with the mRNA vaccines may not be a protective mechanism; rather, it constitutes an immune tolerance mechanism to the spike protein that could promote unopposed SARS-CoV2 infection and replication by suppressing natural antiviral responses.

And here:

Increased IgG4 synthesis due to repeated mRNA vaccination with high antigen concentrations may also cause autoimmune diseases, and promote cancer growth and autoimmune myocarditis in susceptible individuals

They are making exact same point that many other vaccines do not produce sterilizing immunity either. So they are saying that this part of the mRNA vaccine is a normal behavior.

Yes, I'm suggesting that that their reason for emphasizing that (true) point may be in order to proactively counter some misinformation that is/was being spread.

Why do we need to guess their reason? Is the paper factually True or Not? It needs to be confirmed by other studies, for sure. But it raises pretty alarming possibilities.
